Effect of Sodium Lignate Immersion on seed Vigor and Physiological Characteristics of Platycladus orientalis seeds

摘要: 通过人工老化制备出高活力(未经老化)、中活力(老化4d)及低活力(老化7d)的侧柏种子,使用0、25、50、100、200 mg/L木素酸钠溶液浸种处理后,测定种子发芽指标、幼苗生长指标及种子丙二醛(MDA)含量、可溶性蛋白(SP)含量、可溶性糖(SS)含量、脯氨酸(Pro)含量、外渗液电导率(REC)、过氧化物酶(POD)、过氧化氢酶(CAT)活性等生理指标,并对以上指标进行相关性和主成分分析,研究木素酸钠(BCOL)浸种对侧柏种子活力和生理特性的影响结果表明:适宜浓度的BCOL浸种可抑制侧柏种子REC与MDA含量的升高,促进POD、CAT活性增强及SP、SS、Pro累积;高活力和中活力侧柏种子在50 mg/L BCOL处理下相关指标表现最优,低活力侧柏种子在100 mg/L BCOL处理下相关指标表现最优。Abstract: To investigate the effects of sodium lignosulfonate (BCOL) seed soaking on seed vigor and physiological characteristics of Platycladus orientalis, artificial aging was used to prepare P. orientalis seeds with different vigor levels: high vigor (unaged), medium vigor (aged for 4 d), and low vigor (aged for 7 d). The seeds were soaked in BCOL solutions at concentrations of 0, 25, 50, 100, and 200 mg/L, followed by the determination of seed germination indices, seedling growth indices, and physiological indices including malondialdehyde (MDA) content, soluble protein (SP) content, soluble sugar (SS) content, proline (Pro) content, relative electrical conductivity (REC) of exudate, peroxidase (POD) activity, and catalase (CAT) activity. Correlation analysis and principal component analysis were performed on the above indices. The results showed that seed soaking with appropriate concentrations of BCOL could inhibit the increase in REC and MDA content of P. orientalis seeds, promote the enhancement of POD and CAT activities, and facilitate the accumulation of SP, SS, and Pro. Principal component analysis indicated that for high and medium vigor P. orientalis seeds, treatment with 50 mg/L BCOL exhibited the optimal performance in related indices; while for low vigor seeds, the optimal treatment was 100 mg/L BCOL.
