新疆野苹果内生真菌对叶斑病菌的拮抗效果
Inhibitory effects of endophytic fungi isolated from leaves of Malus sieversii on Alternaria alternata via dual culture assay

摘要: 为筛选对新疆野苹果叶斑病菌具拮抗作用的内生真菌资源,以新疆伊犁巩留野果林健康新疆野苹果叶片为材料,采用组织分离法分离可培养内生真菌,结合形态学与ITS序列分析对分离菌株进行分类鉴定,并通过平板对峙法测定内生真菌对叶斑病病原菌链格孢的抑制效果。结果表明:共分离纯化获得内生真菌25株,鉴定为2门4纲7目8科10属,其中子囊菌门为绝对优势类群,占比80%。平板对峙试验显示,9株内生真菌对链格孢的抑制率超过50%,抑菌效果存在显著差异;炭角菌属E01菌株抑菌活性最强,抑制率达64.35%。研究筛选获得的炭角菌属(等拮抗内生真菌,可为新疆野苹果叶斑病菌的绿色生物防治提供菌种资源。Abstract: To screen endophytic fungal resources with antagonistic effects against leaf spot disease of Malus sieversii, healthy leaves of Malus sieversii were collected from the Gongliu Wild Fruit Forest in Yili, Xinjiang. Culturable endophytic fungi were isolated using the tissue isolation method, and identified by combining morphological characteristics and ITS sequence analysis. The inhibitory effects of endophytic fungi against Alternaria alternata, the pathogen causing leaf spot disease, were determined by the plate confrontation assay. The results showed that a total of 25
1 endophytic fungal strains were isolated and purified, which were identified as 10 genera, 8 families, 7 orders, 4 classes and 2 phyla. Among them, Ascomycota was the absolutely dominant phylum, accounting for 84.6%. The plate confrontation test indicated that 9 endophytic fungal strains exhibited an inhibition rate higher than 50% against A. alternata, with significant differences in the inhibitory effects. Strain E01 of Xylaria showed the strongest antifungal activity, with an inhibition rate of 64.35%. The antagonistic endophytic fungi obtained in this study, such as Xylaria , provide strain resources for the green biological control of Alternaria alternata in Malus sieversii.
