橡胶树HbWRKY12a调控拟南芥开花时间及盐胁迫响应

摘要: 为阐明橡胶树WRKY转录因子的生物学功能,利用生物信息学方法进行HbWRKY12a的系统发育分析,通过RT-PCR扩增获得HbWRKY12a基因片段并完成基因克隆,采用异源过表达策略,在拟南芥中构建HbWRKY12a过表达转基因材料,并对其开花时间及盐胁迫响应进行分析。结果表明:与野生型相比,HbWRKY12a-OE过表达植株在长日照和短日照条件下均表现出显著提前开花的表型;在盐胁迫处理条件下,过表达植株的耐盐性显著降低。这些结果表明橡胶树HbWRKY12a参与调控植物开花时间及盐胁迫响应过程。Abstract: WRKY transcription factors play critical roles in plant development and stress responses. To elucidate the biological function of the WRKY transcription factor in rubber tree (Hevea brasiliensis (Willd. ex A. Juss.) Müll. Arg.), bioinformatics methods were used to conduct phylogenetic analysis of HbWRKY12a. The gene fragment of HbWRKY12a was amplified and cloned via RT-PCR. HbWRKY12a was heterologously overexpressed in Arabidopsis thaliana, and its effects on flowering time and salt stress responses were examined. The results showed that compared with wild-type plants, HbWRKY12a overexpression lines exhibited significantly earlier flowering under both long-day and short-day conditions. Moreover, HbWRKY12a overexpression resulted in a marked reduction in salt tolerance under salt stress. These findings indicate that HbWRKY12a functions as a regulator of flowering time and salt stress responses in plants.
