豹皮樟扦插生根特性研究

Study on Rooting Characteristics of Litsea coreana Levl var. sinensis Cuttings

  • 摘要: 为探究木质化程度与激素浓度对豹皮樟嫩枝扦插生根的影响,采用双因素完全随机区组设计,分析插穗木质化程度与IBA·NAA浓度对插穗根系形态及生理代谢的影响,并通过解剖学研究,明确豹皮樟扦插生根特性。结果表明:低木质化插穗生根率显著高于半木质化插穗,IBA·NAA能显著提升扦插生根率与根系形态指标;最佳处理组合为100 mg/L的IBA·NAA处理低木质化插穗,培养120 d后平均生根率达90%;豹皮樟属诱导生根类型,生根方式可分为皮部生根型与混合生根型,根原基主要发生于中柱鞘,部分起源于愈伤组织、韧皮部及薄壁细胞等组织;IBA·NAA有助于提高插穗可溶性糖、蛋白含量,增加吲哚乙酸氧化酶、过氧化物酶和多酚氧化酶酶活性,刺激生长素与细胞分裂素含量升高并降低总酚含量,以此维持插穗生理活性。

     

    Abstract: To investigate the effects of lignification degree and hormone concentration on the rooting of Litsea coreana Levl var. sinensis softwood cuttings, a two-factor completely randomized block design was employed to analyze the influences of cutting lignification degree and IBA·NAA concentration on root morphology and physiological metabolism. Additionally, anatomical studies were conducted to clarify the rooting characteristics of L. coreana var. sinensis cuttings. The results showed that the rooting rate of cuttings with a low degree of lignification was higher than that of semi-lignified cuttings. IBA·NAA significantly improved the rooting rate and root morphological indices of the cuttings. The optimal treatment combination was 100 mg/L IBA·NAA applied to low-lignification cuttings, which achieved an average rooting rate of 90% after 120 days of cultivation. L. coreana var. sinensis exhibits an induced rooting type, and the rooting pattern can be classified into Cortex rooting type and mixed rooting type. Root primordia mainly originate from the pericycle, with partial origins from callus, phloem, parenchyma cells, and other tissues. IBA·NAA helps to increase the contents of soluble sugars and proteins in cuttings, enhance the activities of indoleacetic acid oxidase (IAAO), peroxidase (POD), and polyphenol oxidase (PPO), stimulate the increase in auxin and cytokinin levels, and reduce the total phenol content, thereby maintaining the physiological activity of the cuttings.
