微波处理对老化侧柏种子活力和生理特性的影响
Effects of Microwave Treatment on Vigor and Physiological Characteristics of Aged Platycladus orientalis Seeds

摘要: 为了解决侧柏种子老化、活力不强等问题,以侧柏种子为材料,采用高温高湿法对种子进行人工加速老化处理,测定不同老化处理时间下侧柏种子的发芽力,并采用300 W、500 W、600 W(分别记为W3、W5、W6)3种功率和10 s、20 s、40 s、60 s(分别记为T1、T2、T4、T6)4种时间的组合对老化4 d的侧柏种子进行微波处理,测定种子发芽率、相对电导率、超氧化物歧化酶(SOD)活性、过氧化物酶(POD)活性及丙二醛(MDA)、可溶性糖(SS)、可溶性蛋白(SP)、三磷酸腺苷(ATP)含量等指标,并运用隶属函数法对不同微波处理组合进行综合评价。结果表明:侧柏种子老化处理后,其发芽率、发芽势、发芽指数、活力指数显著下降,老化4 d种子发芽率从86.67%降至69.67%。随着微波功率和时间的增加,老化侧柏种子的发芽率、抗氧化酶、贮藏营养物质和ATP含量呈先增加后降低趋势,相对电导率和MDA含量呈先降低后增加趋势。其中300 W 40 s(W3T4)处理发芽率、抗氧化酶、贮藏营养物质与ATP含量显著高于CK,相对电导率与MDA含量显著低于CK。高功率600W长时间(W6T6)微波处理对种子萌发有抑制作用。因此,适宜的微波处理对老化侧柏种子萌发有促进作用,其中300 W 40s处理效果最好。Abstract: To solve the problems of aging and low vigor of Platycladus orientalis seeds, this study used P. orientalis seeds subjected to artificial accelerated aging via high-temperature andhigh-humidity methods. Germination indices of P. orientalis seeds under different aging durati-ons were measured. Aged seeds (4 days) were then treated with microwave at three power l-evels (300 W, 500 W, 600 W; denoted as W3, W5, W6) and four exposure times (10 s, 20 s, 40 s, 60 s; denoted as T1, T2, T4, T6). The following parameters were evaluated: germin-ation percentage, relative electrical conductivity, malondialdehyde (MDA) content, soluble sug-ar (SS) content, soluble protein (SP) content, superoxide dismutase (SOD) activity, peroxidase (POD) activity, and adenosine triphosphate (ATP) content. A comprehensive evaluation of diff-erent microwave treatments was conducted using the membership function method. The resultsshowed that after aging treatment, the germination percentage, germination potential, germinati-on index, and vigor index of P. orientalis seeds significantly decreased. The germination perc-entage of seeds aged for 4 days declined from 86.67% to 69.67%. As microwave power and exposure duration increased, the germination percentage, antioxidant enzyme activities, reserve nutrient content, and ATP levels of aged P. orientalis seeds exhibited an initial increase follo-wed by a subsequent decrease. Conversely, relative electrical conductivity and malondialdehyde(MDA) content demonstrated an initial decline followed by a rise. Among all treatments, the 300 W for 40 s (W3T4) protocol significantly enhanced germination percentage, antioxidant e-nzyme activities, reserve nutrients, and ATP content compared to the control, while simultane-ously reducing relative electrical conductivity and MDA levels. Prolonged exposure to high-p-ower microwave treatment (W6T6) exerted inhibitory effects on seed germination.Comprehensi-ve analysis indicated that appropriate microwave treatment promotes the germination of aged P. orientalis seeds, with the 300 W 40 s (W3T4) treatment demonstrating the most effective results.
