Establishment of Regeneration System from Hypocotyls of Castanea mollissima cv.'Yanshanhong'

Study on tissue culture of Castanea mollissima cv.'Yanshanhong' was carried out with hypocotyls as explant.The results showed that the best primary culture was obtained by inoculating hypocotyl explant on the medium of MS+6-BA 2.0 mg/L+sucrose 20 g/L+agar 7 g/L after 13~15 min of surface disinfection in 0.1% Hg solution.According to the result of quadratic regressive orthogonal device, the best subculture medium was WPM+6-BA 1.25~1.53 mg/L+IBA 0.08~0.15 mg/L+sucrose 30 g/L+agar 7 g/L.It was showed that the indirect rooting method in darkness on medium of 1/4 MS+IBA 3.0 mg/L for 4 days and then transferred to bare medium of 1/4 MS could get better rooting results than the direct rooting method with medium of 1/4 MS+IBA 1.0 mg/L, The smaller calluses and stronger roots could be produced by the indirect rooting method.
