Establishment of Efficient Propagation System for 2 Chrysanthemum Cultivars

  • Tissue culture experiment was conducted for 2 chrysanthemum cultivars 'pudijin' and 'beilinhong' to acquire plantlets by adventitious bud differentiation from leaf explant on MS medium by applying different concentrations and proportions of 6-BA and NAA plant growth regulators. The results showed that MS+6-BA 2.0 mg/L+NAA 1.0 mg/L was the optimal leaf propagation medium for 'Pudijin' cultivar, on which the leaf proliferation rate was 96%, while the petiole proliferation rate was 90%. The optimal leaf propagation medium for 'Beilinhong' cultivar was MS+6-BA 1 mg/L+NAA 0.5 mg/L, on which the proliferation rate was up to 96%, but other explants of the cultivar such as petiole or stem could not be propagated on this medium. The optimal axillary bud propagation medium for 'Pudijin' cultivar was MS+6-BA 0.5 mg/L+NAA 0.01 mg/L, by which the mean number of adventitious bud propagated from each axillary bud reached 5.5. MS+6-BA 0.5 mg/L+NAA 0.05 mg/L and MS+6-BA 0.5 mg/L+NAA 0.01 mg/L were the optimal propagation media for axillary bud explant of 'Beilinhong' cultivar, on which the average numbers of 6.3 and 5.6 adventitious buds could be obtained respectively from each axillary bud, while there was no significant difference between the number of adventitious buds obtained on the 2 media.
