Studies on Callus Culture of Taxus chinensis var. mairei

  • The induction condition and optimal medium constitution for callus growth from leaf and stem explants of Taxus chinensis var. mairei were studied. The results indicated that the callus induction frequency from young stems and leaves on the MS medium with 2.0 mg/L of 2,4-D was higher than those on the MS medium with 0.5 mg/L or 1.0 mg/L 2,4-D. The inducing frequency of callus from young stems varied with the combination of different plant growth regulators and different concentrations of the plant growth regulators. Compared to other combinations, the callus induction frequency from young stems as explants reached 100%, 90%, 75% and 75% respectively on the media MS+2,4-D 2.0 mg/L, MS+2,4-D 2.0 mg/L+6-BA 1.0 mg/L, MS+6-BA 1.0 mg/L+IAA 1.0 mg/L and 1/2 MS+NAA 2.0 mg/L+IBA 1.0 mg/L.
