Genetic Diversity Analysis of Wild Paphiopedilum Species in the South of Guizhou

  • SRAP marker was used to detect the genetic diversity of 8 accessions of wild Paphiopedilum species. 8 pairs of polymorphic and repeatable primers were screened out for amplification and establishing the optimal PCR reaction system and genetic relationships of wild Paphiopedilum were detected by UPGMA cluster analysis. Totally 152 clear bands were revealed, including 122 polymorphic bands, and the percentage of polymorphic bands was 80.26%. The mean observed number of alleles (Na) was 1.763 2, the effective number of alleles (Ne) was 1.506 1, the Nei′s gene diversity index (H) was 0.287 9 and the Shannon information diversity index (I) was 0.424 1. The genetic identity coefficient was between 0.513 to 0.756. Divided wild Paphiopedilum from different sources into 3 groups with the threshold value of identity coefficient as 0.67, the study showed that the clustering of Paphiopedilum species based on the SRAP markers was consistent with that from morphological classification.
