Using Puncture Method to Improve the Survival Rate of Shoot-tip Micrografting Technology

  • Experiment with Citrus maxima cv. Mansailong as root stock, the adult stem segments of different C.paradisi cultivars were used as explants to induce adventitious buds, which could improve the survival rate of grafted seedlings. Adventitious buds induced by stem segments at different concentrations of plant growth regulator were used as scions, the effect of different grafting methods, scion varieties and sources on the survival rate of shoot-tip micrografting was studied. The results showed that the best medium for adventitious bud induction is WPM + 6-BA 1.0 mg/L + NAA 0.2 mg/L + GA 0.2 mg/L; it was found that the puncture method was more suitable for C.paradisi shoot-tip micrografting and it can significantly improve the survival rate of grafting; Shoot-tip micrografting has a significant differences in the graft survival rate of different scion varieties C.paradisi, the survival rate was Ray Ruby > Star ruby > Hudson Foster > Flame > Rio Red; Induction of adventitious bud of graft survival rate was 76.67%, field experiment graft survival rate is 38.33%.
