The Effect of Culture Components on Producing Ligninolytic Enzymes of Microbial Consortium FHs and Its Optimization

Abstract
Using microbial consortium FHs (LS136, LJ485, LJ496) as the experimental materials, the significant effects of culture medium on producing ligninolytic enzymes were studied by Plackett-Burman experiment design. Then, using lignin degradation rate as the evaluation standard, the response surface method was used to optimize the culture medium of the FHs, and the lignin degradation rate of single strains were compared with it for analysis. The results showed that the significant affecting factors of producing ligninolytic enzymes in GPD culture medium of FHs were glucose, magnesium sulfate, guaiacol, thiamine, respectively. In addition, the results of response surface optimization showed that the optimal media components of FHs were 4.0 g/L glucose, 1.9 g/L magnesium sulfate, 3.0 mmol/L guaiacol, 70 mg/L thiamine, respectively. With the condition of these, the lignin degradation rate was up to 37.3%, and the effects of lignin degradation rate was increased by 24.1%, 28.0% and 44.0%, respectively, compared with the single strains.
