Adventitious Bud Induction and Rooting Culture of Macadamia ternifolia

  • The shoot segments of macadamia nut cultivar 'H2' were used as explant materials, and WPM, MS and 1/2MS were used as the basic medium to study the effects of different exogenous hormones and their ratios on axillary bud initiation, proliferation and rooting of tissue cultured seedlings. The results show that suitable medium for axillary bud initiation is WPM + 6-BA 2.0 mg/L + IBA 1.0 mg/L + AC 200 mg/L, and the induction coefficient is 3.51. Meanwhile, the sprouts grow robustly after axillary buds germination. The optimal proliferation medium for axillary buds is MS + 6-BA 2.0 mg/L + KT 0.5 mg/L, and the proliferation coefficient reaches 3.49. The optimum rooting formula selected in this study is 1/2MS + 6- BA 0.2 mg/L + IBA 0.5 mg/L + ABA 0.5 mg/L + AC 200 mg/L, the rooting rate is 42.2%.
