Study on the Ratio of Nutrients for Promoting Pollen Germination of Camellia oleifera

Abstract
We study the effects of eight different nutrients maters including the sucrose, boric acid, urea, calcium chloride, potassium dihydrogen phosphate, PEG−3350, vitamin C and vitamin B1 on the pollen germination rate(PGR) and pollen germination length(PGL) of Camellia oleifera under the solid medium. The mature pollen of 'Ganwu2', 'Gan70', 'Changlin 166' were collected randomly, the single factor and complete orthogonal tests were used, to screen the sucrose-boric acid basic medium and other nutrients for promoting the pollen germination of C. oleifera, finally, the best nutrients composition could be determined. The result shows that the basic medium for pollen germination of C. oleifera was 5 g/L AGAR+100 g/L sucrose+0.075 g/L boric acid. KH2PO4 and PEG−3350 had obvious promoting effect on the pollen germination and pollen tube elongation of C. olerifera, while urea and calcium chloride without significant promoting effect. Vitamin C and Vitamin B1 had certain inhibitory effect on the pollen germination rate of C. oleifera, but they promoted the elongation of pollen tube. The best matters for promoting the pollen germination of C. oleifera were KH2PO4 (40–120 mg/L) and PEG−3350 (50–200 mg/L). The optimal nutrients composition for promoting the pollen germination of Camellia oleifera was as follows: The basic medium(5 g/L AGAR+100 g/L sucrose+0.075 g/L boric acid)+60 mg/L KH2PO4+150 mg/L PEG−3350.
