Effects of Cinnamomum glanduliferum matrix addition on the growth characteristics of Antrodia cinnamomea

  • ObjectiveTo explore the effect of Cinnamomum glanduliferum substrate addition on the growth and physiological and biochemical characteristics of Antrodia cinnamomea.
    MethodPotato agar medium (PDA) was supplemented with Ten concentrations (0.125-64 g/L) of C. glanduliferum branches, leaves and their mixtures and cultured at constant temperature for 30 days, and the differences in colony diameter, mycelial density, colony color, colony growth potential, mycelial growth rate, growth index, biomass, superoxide dismutase (SOD) activity, and total triterpene (TT) content were analyzed.
    ResultUnder the condition of adding C. glanduliferum branches in PDA medium, the growth characteristics and biomass of A. cinnamomea were higher when the addition concentration was 1-8 g/L, and the SOD activity and TT content of A. cinnamomea were significantly higher when the addition concentration was 2-4 g/L and 2 g/L, respectively; Under the condition of adding young leaves of C. glanduliferum to PDA medium, its growth characteristics and biomass were higher when the concentration was 1-4 g/L and 1-16 g/L, respectively. When the concentration was 0.5-2 g/L, its SOD activity and TT content were significantly higher than those of the control; The growth characteristics, biomass, SOD activity and TT content were kept at a high level when the addition concentration was 2-16 g/L, 4-16 g/L, 16-32 g/L and 8-32 g/L, respectively, when the branches and leaves mixture of C. glanduliferum was added to the PDA medium.
    ConclusionAdding 4 g/L of C. glanduliferum young leaf as the substrate has the best promoting effect on the growth indicators and biomass of A. cinnamomea, with a colony diameter of 83.40 mm, a mycelial growth rate of 2.78 mm/d, and a biomass of 0.23 g; Adding 2 g/L of C. glanduliferum branches matrix significantly increased the SOD activity of A. cinnamomea (392.06 U/g); The addition of 16 g/L mixed substrate of C. glanduliferum branches and leaves significantly promoted the total triterpene content of A. cinnamomea (29.32 mg/g). This study can provide theoretical and practical basis for the optimization of artificial cultivation conditions and large-scale cultivation of A. cinnamomea.
