The Factors Influencing the Embryogenic Callus Induction of Litsea cubeba

  • In order to screen out the best induction conditions for embryogenic calls formation of Litsea cubeba, nearly mature seed embryos of L. cubeba good single plants were used as explants to analyze the effects of different disinfection methods, hormone combinations, carbon source type and seed source type on the embryogenic calls induction. The results showed that sterilizing the endocarp with 75% ethanol for 5 min and rinsing with sterile water for 5 times resulted in the lowest contamination rate of 0.00%; Nanchang City, Jiangxi Province, was the best seed source location for embryogenic calls induction, with an induction rate of 7.04%; and the addition of the exogenous hormone 1.0 mg/L 6−BA + 0.2 mg/L 2,4−D + 0.5 mg/L IAA had the highest rate of embryogenic calls, with a total induction rate of 17.50%. In this study, we successfully induced embryogenic calls of L. cubeba and achieved proliferation of embryogenic calls and differentiation of somatic embryos in medium MS + 0.5 mg/L 6−BA + 0.1 mg/L 2,4−D + 0.25 mg/L IAA, and then realized the germination of somatic embryos.
