Study on Optimization of Magnolol Extraction by Orthogonal Design and HPLC Analysis

  • The schemes for extracting phenolic materials from Magnolia officinalis were studied by orthogonal design under the condition of water bath and ultrasonic treatment. The results showed that the optimal scheme to extract magnolol and honokiol was as follows. The chromatographic conditions included that the SinoChrom ODSBP (4.6mm×150mm, 5μm) was used as chromatographic column by HPLC isolation; the methanol water (78∶22) as mobile phase; the flow rate was 1.0mL/min, the detection wavelength was 294nm, the column temperature was 30℃ and the injection volume was 20μL. The highest extracting rate for the phenolic materials and the best separation effect were obtained under the conditions of 80% ethanol as the reagent with 20min ultrasonic treatment, soaking for 50min at 50℃.
