Isolation of Cellulose Degradating Bacteria from Bark BeetlesIntestine and Enzyme Production Properties

  • Five strains of cellulose degrading bacteria were isolated from the intestine of young bark beetle, and the activity of cellulase generated by these strains was determined by Dus cellulose selective medium on filter paper or absorbent cotton with CMC-Na as fermentation substrate. The results showed that HS-11-5 was the strain with the highest cellulase activity (32.62 U/mL), whose optimum cellulase producing conditions were as follows: 50h of fermentation; (NH4)2SO4 as nitrogen source; under pH value of 7.5-8.5 at 28-30℃; and 12%-16% as the medium loading proportion.
