五针白皮松离体胚培养初步研究

A STUDY ON THE EXCISED EMBRYO CULTURE OF PINUS SFUAMATA

  • 摘要: 对珍稀濒危植物五针白皮松进行离体胚的组织培养,可以从子叶上诱导产生大量的愈伤组织及不定芽。经初步试验,以Ms为基本培养基并附加激素BA3mg/L,NAA0.05mg/L,对诱导子叶产生不定芽最为有效,而Ms+BA1~3mg/L+NAA0.05~0.5 mg/L可诱导培养的离体胚产生愈伤组织。胚根及胚轴的下部分在培养中逐渐死亡。

     

    Abstract: Numerous adventitious buds and calli were induced from cotyledons by culturing mature embryos of Pinus sfuamata on Ms medium supplemented with BA and NAA.The results showed that medium of Ms+BA 3×10-6 +NAA0.05×10-6 was valid for inducing adventitious buds, and the calli could be brought out from cotyledons cultured on Ms medium supplemented with BA 1~3mg/L and NAA 0.05~0.5mg/L.The radicles and the lower part of the plumular axis perished gradually.
