西南桦组织培养研究

A Study on Tissue Culture of Betula alnoides

  • 摘要: 对西南桦组培外植体和培养基的选取、组培方法的筛选进行了研究,并培育出了西南桦的组培植株.在此基础上,分析了西南桦组织培养过程中愈伤组织的诱导和胚状体形成过程,并对其影响因子进行了分析:认为春、秋两季的材料最易产生愈伤组织;6BA/NAA的比值越大,分化出的不定芽就越多;黑暗和光照条件下均可从胚性愈伤组织中诱导出胚状体;含5mg/LABA和20g/L蔗糖的MS培养基最适于诱导西南桦体细胞胚分化.

     

    Abstract: Selection of explants and culture mediums and methods of tissue culture in Betula alnoides are studied.Regenerated plants from tissue culture are obtained.Based on the above study, program of callus initiation and forming embryoid during tissue culture are analyzed.Meanwhile, affecting factors of Tissue culture to Betula alnoides also are analyzed as follows:The explant selected either in spring or in autumn are the easiest to produce callus;the bigger the ratio of 6BA to NAA is, the more adventitious buds are;the embryoid could be induced either in dark or light conditions from embryoid callus;the most suitable medium for producing the somatic embryoid differention is the MS medium which contains 5mg/L ABA and 20g/L sucrose.
