Abstract:
The influence upon callus induction and its growth by the category and size of the explants, method of implanting, combination of plant growth regulators, and the effect of AgNO
3 and vitamin C was studied when the segments of stem, shoot, leaf and root of Aloe barbadensis were applied as explants, MS was adopted as elementary medium added with varied quantities of 2, 4-D, NAA, KT and the different combinations of these plant growth regulators, and different contents of AgNO
3 and Vitamin C were added into the medium.It was indicated by the results that the best explants were 2 cm long stems, the best implanting method was the horizontal placement, the best combination of plant growth regulators was NAA 15 mg/L+2, 4-D 4 mg/L+KT 0.1 mg/L, with which the callus induction rate reached to be 90%.It was also showed that the addition of 10 mg/L AgNO
3 could promote the growth of the callus, and 5 mg/L of vitamin C was helpful to inhibit callus browning in the culture process.
