燕山红栗下胚轴再生体系的建立

Establishment of Regeneration System from Hypocotyls of Castanea mollissima cv.'Yanshanhong'

  • 摘要: 以燕山红栗的下胚轴为外植体,进行组织培养研究,结果表明:用0.1%升汞表面灭菌13~15 m in后,以平卧的方式接种在MS+6-BA 2.0 mg/L+蔗糖20 g/L+琼脂7 g/L的培养基上进行初代培养效果最佳;通过二次旋转回归正交试验得出,最适继代培养基为WPM+6-BA 1.25~1.53 mg/L+IBA 0.08~0.15 mg/L+蔗糖30 g/L+琼脂7 g/L;采用间接生根法即在黑暗条件下用1/4MS+3.0 mg/L IBA诱导生根4 d,后转入空白培养基上,比直接在添加IBA 1.0 mg/L生根培养基中进行诱导生根效果好,间接诱导生根不仅愈伤组织小,而且根粗壮。

     

    Abstract: Study on tissue culture of Castanea mollissima cv.'Yanshanhong' was carried out with hypocotyls as explant.The results showed that the best primary culture was obtained by inoculating hypocotyl explant on the medium of MS+6-BA 2.0 mg/L+sucrose 20 g/L+agar 7 g/L after 13~15 min of surface disinfection in 0.1% Hg solution.According to the result of quadratic regressive orthogonal device, the best subculture medium was WPM+6-BA 1.25~1.53 mg/L+IBA 0.08~0.15 mg/L+sucrose 30 g/L+agar 7 g/L.It was showed that the indirect rooting method in darkness on medium of 1/4 MS+IBA 3.0 mg/L for 4 days and then transferred to bare medium of 1/4 MS could get better rooting results than the direct rooting method with medium of 1/4 MS+IBA 1.0 mg/L, The smaller calluses and stronger roots could be produced by the indirect rooting method.
