用rep-PCR方法研究细菌遗传多样性的探讨

Studies on Genomic Diversity of Bacteria by rep-PCR

  • 摘要: 对大肠杆菌、枯草杆菌、四联球菌、普通变形杆菌、产气肠杆菌基因组DNA进行rep-PCR扩增,并在模板样品中混入植物DNA作对照实验.结果表明:用rep-PCR方法分析细菌基因多样性,能在较大范围内避免植物基因的干扰.模板量梯度实验结果显示:扩增产物多态性与模板量相关.

     

    Abstract: The genomic DNAs of Escherichia coli,Bacillus subtilis,Micrococcus tetragenus,Proteus vulgaris and Enterobacter aerogenes were analyzed by rep-PCR with BOX primers,and the CK experiment was carried out by adding plant genomic DNAs into the bacteria genomic DNAs.It was showed that the rep-PCR with BOX primers could identify these bacteria and the bacteria polymorphism was not interfered by plant genomic DNAs in a large range of DNA template concentration.It was also showed by the results from the gradient experiment with template quantities that the bacteria rep-PCR polymorphism was related to template concentration.
