扫描电镜技术在动物毛发微观结构研究上的应用

Application of Scanning Electron Microscope to Study on Microscopic Structure of Animal Hair

  • 摘要: 用6种不同的扫描电镜样品制备方法对动物毛发表面及横断面的超微结构进行观察比较.结果表明:正常、清洁的动物毛发采用常规方法即可得到真实、清晰的超微结构,而脏发和受损毛发,必须进行适当的清洗处理,有效除去覆盖物,真实结构才能完全显示出来.6种方法中用洗涤剂先清洗、无水丙酮浸泡4 h,结合超声波清洗的方法清洗毛发样品效果最好.

     

    Abstract: The surface and cross section of animal hairs were observed and compared by different methods of SEM specimen preparation.The results showed that real and vivid ultrastructure could be observed by using routine means for normal and clean hairs.While dirty and damaged hairs must be cleaned firstly to remove slipcover,then real structure could be showed.Of six kinds of specimen preparation methods,the best one was that hairs were firstly washed with detergent and then soaked in acetone for 4 hours combined with ultrasonic cleaning before being observed.
