Abstract:
The endophytic fungal strains isolated from different ages of
Platycladus orientalis in Huangdi Mausoleum of Huangling County in Shaanxi Province were identified by tissue isolation method combined with molecular biological data and morphological characteristics. The endophytic fungi showed the highest isolation rate of 53.2% at the age less than 100 years. While the age greater than or equal to 100 years with the age less than 600 years, and the age greater than or equal to 600 years, the separation rate was lower, 24.6% and 24.8% respectively. The endophytic fungi of different age stages showed rich diversity, and the index was between 2.4 and 2.7. The similarity of endophytic fungi population was different in different age stages. The similarity coefficient of endophytic fungi between the age greater than or equal to 100 years with less than 600 years and greater than or equal to 600 years was 0.743; The similarity between the age less than 100 years and the age greater than or equal to 100 years with less than 600 years was 0.526; The similarity between the age less than 100 years and the age greater than or equal to 600 years was 0.541. Dominant species of
Dimorphospora, Epicoccum, Alternaria, and
Paecilomyces were the predominant microflora of all age groups.
