滇牡丹根皮初提物抗菌活性研究

Antimicrobial Activity from the Root Bark of Paeonia delavayi Original Extract

  • 摘要: 采用乙醇与丙酮分别提取滇牡丹根皮内物质,用13种致病细菌进行活性筛选,采用抑菌圈法评价相应初提物的抗菌活性,并测定具有抗菌活性初提物的最低抑制浓度(MIC),并检测不同温度和不同pH处理后滇牡丹根皮中抗菌物质的抑菌效果。结果表明:乙醇初提物与丙酮初提物能够对供试13种致病细菌中的5种致病细菌具有明显抑菌效果,最低抑制浓度基本相同,只有对无乳链球菌与副溶血弧球菌的MIC值不同;滇牡丹根皮乙醇初提物在不同温度与不同pH处理后,各个处理部分抑菌圈大小没有出现明显变化,因此滇牡丹根皮乙醇初提物有较好的热稳定性和酸碱稳定性;滇牡丹乙醇初提物经过大孔树脂分段纯化后,发现只有100%乙醇洗脱部分具有抗菌活性。

     

    Abstract: Primary extract were extracted from roots of Paeonia delavayi by ethanol and acetone. And then the activity of the extract was screened by 13 kinds of pathogenic bacteria. Inoculating inhibition zone was used to evaluate the antibacterial activity of primary extract and minimum inhibitory concentration (MIC) were determined. Stabilization of ethanol extracts from roots of P.delavayi was detected by treating with different temperature and different pH. The result showed the ethanol extracts and acetone extracts had inhibitory effect to the 5 pathogenic bacteria of 13 pathogenic bacteria, the MIC of 2 extracts to Bacillus cereus, Straphylococcus haemolyticus, Escherichia coli were similar, and MIC to Streptcococcus agalactiae and Vibrio parahaemolyticus had slight difference. The antibacterial activity of ethanol extracts treated with different temperatures and different pH did not change significantly. It means that stability of the ethanol extracts from roots of P.delavayi was good. After macroporous resin treatment, only 100% ethanol eluted fraction showed antibacterial activity.
