基于扫描仪和ImageJ软件的香樟种子测量与种源分析

Seed Size Measurement and Provenance Analysis of Cinnamomum camphora Based on Scanner and ImageJ

  • 摘要: 利用扫描仪获取标准图形, 结合ImageJ软件测定香樟种子大小, 通过与游标卡尺法进行比较和相关性分析, 利用该方法对江西境内具有代表性的10个种源香樟种子大小进行了测量, 并分析了其种源及家系间的变异水平。结果表明:2种方法测量的种子大小呈显著正相关(P < 0.05), 相关系数大于0.986 8;种子大小之间的变异主要来自于种源间, 群体内种子大小的遗传多样性水平较低, 平均多样性指数为1.05, 且受地理气候因子影响较小; 相比游标卡尺法, 图像测定法能快速、准确测定大批量香樟种子大小。

     

    Abstract: This research aimed at measuring Cinnamomum camphora seed size by the scanner which obtained standard graphics and ImageJ software which analyzed the seed size.Comparison and correlation analysis were based on vernier caliper method.Variation between the provenance and family level of seed size of 10 typical species of C.camphora was analyzed by this method.The results showed the correlation coefficient was larger than 0.986 8 which indicated the two methods were significantly positively related (P < 0.05).C.camphora seed size variation mainly from the provenance.The average diversity index was 1.05 which suggested small genetic variation level among populations, and less affected by climatic factors.Comparing with vernier caliper method, image determination was more rapid and accurate in determining C.camphora seed size.
