响应面法优化云南松树皮多聚原花青素片段化条件

Optimization for Fragmentation Process of Polymeric Proanthocyanidins from Pinus yunnanensis Barks by Response Surface Methodology

  • 摘要: 以云南松树皮PPC为原料,片段化产物1的含量为评价指标,通过单因素试验分析了反应温度、反应时间、盐酸浓度和原料/茶多酚比(PPC/TP)对云南松树皮多聚原花青素片段化主产物生成的影响,采用响应面法优化其反应条件,以主产物含量为响应值,对30个试验点调查数据进行多元非线性分析,并通过方差分析对回归模型进行显著性检验。结果表明,最佳片段化条件为反应温度51 ℃、反应时间120 min、盐酸浓度3%、原料/茶多酚质量比为1:1。在此条件下,片段化主产物的含量达到861.897 μg/mL,与理论预测值(864.664 μg/mL)较为接近,说明响应面法优化云南松树皮多聚原花青素片段化反应方法可行。

     

    Abstract: Taking PPC of Pinus yunnanensis bark as material, the content of fragmented product 1 was the evaluation index. The effects of reaction temperature, reaction time, concentration of hydrochloric acid and PPC/TP on the formation of fragmented proanthocyanidins from P.yunnanensis bark were analyzed by single factor experiments. Response surface method was used to optimize the reaction conditions, the main product content as a response value, the survey data of 30 test points were subjected to multivariate nonlinear analysis, and a significant test of regression model was conducted by ANOVA. The results showed that the best fragmentation conditions were reaction temperature 51 ℃, reaction time 120 min, HCl concentration 3% and PPC/TP ratio=1:1. Under these conditions, the actual content of the product reached 861.897 μg/mL that well matched with the predicted values (864.664 μg/mL), indicated it was feasible that the response surface method to optimize the fragmentation process of PPC from P.yunnanensis barks.
