荷花转录组测序及花青素苷合成相关基因表达分析

Transcriptome Sequencing and Expression Analysis of Anthocyanin Synthesis Related Genes in Lotus

  • 摘要: 以荷花不同花色品种‘青玉’和‘白洋淀红莲’为研究对象,运用高通量测序技术,对松蕾期的花瓣进行转录组测序,并对2个测序文库进行差异表达及实时荧光定量PCR分析。结果表明:共获得1 142条差异表达基因。筛选出与花青素苷合成相关的关键基因ANSCHSDFRUF3GT 。4个基因在两种荷花花瓣不同时期中的表达量存在显著差异,在松蕾期或初花期的表达量最高。

     

    Abstract: In this study, different varieties of lotus ′Qingyu′ and ′Baiyangdianhonglian′ were selected as the research materials. The transcriptional sequence of petals at bud stage was sequenced by high-throughput sequencing technology. A total of 1 142 DEGs were identified from the 2 sample sequencing libraries, and the main genes of anthocyanin biosynthesis were screened out including ANS genes, CHS genes, DFR genes and UF3GT genes. Real-time PCR expression analysis showed that there was significant difference in the expression of 4 genes in the 2 kinds of flower petals at different stages. The highest expression level was at the bud stage or early flowering stage. This study lays the foundation for further study on the molecular mechanism of anthocyanin biosynthesis and flower color breeding.
