2株楚雄腮扁叶蜂虫生真菌培养条件及产孢特性研究

The Culture Conditions and Sporulation Characteristics of 2 Entomopathogenic Fungi with Cephlica chuxiongica

  • 摘要: 对2株楚雄腮扁叶蜂虫生真菌(NH512、Y512)菌株培养条件及产孢特性进行研究,分析温度、光照及紫外线诱导对菌株生长和产孢的影响。结果表明:NH512菌株菌丝的生长最适温度为25 ℃,在此温度下培养15 d时的平均菌落直径为48.15 mm;Y512菌株菌丝的生长最适温度为20 ℃,在此温度下培养15 d时的平均菌落直径为64.20 mm;25 ℃条件下培养15 d,NH512菌株和Y512菌株累计产孢量均最大,分别为9.184 × 107个/cm2和3.091 × 107个/cm2。黑暗-光照12 h交替条件可以促进NH512菌株和Y512菌株菌丝生长,培养15 d时平均菌落直径均最大,分别为40.10、44.40 mm;无光照条件有利于NH512菌株产孢,而黑暗-光照12 h交替条件能促进Y512菌株产孢,培养15 d时NH512菌株、Y512菌株平均产孢量分别为9.184 × 107、9.038 × 107个/cm2。紫外线诱导能促进NH512菌株的菌丝生长,抑制Y512菌株的菌丝生长;紫外线诱导能抑制NH512菌株产孢,而紫外线诱导能大大促进Y512菌株产孢,平均累计产孢量增加到15.69 × 107个/cm2

     

    Abstract: In the study, the culture conditions and sporulation characteristics of 2 entomopathogenic fungi (NH512, Y512) of Cephlica chuxiongica were studied, and the effects of temperature, light and UV mutagenesis on the growth and sporulation of fungi were analyzed. The results showed that the optimum temperature for mycelial growth of the strain NH512 is 25 ℃, in which the average colony diameter is 48.15 mm on the 15 day. The optimum temperature for mycelial growth of the strain Y512 is 20 ℃, in which the average colony diameter is 64.20 mm on the 15 day. Under the 25 ℃ condition of cultivation on the 15 day, the accumulation of spores of the strains NH512 and Y512 are maximum, which are 9.184 × 107/cm2 and 3.091 × 107/cm2, respectively. Both the alternating conditions of dark and light for 12 hours can promote the growth of 2 strains, and the average colony diameter are the maximum on the 15 day, which are 40.10 mm and 44.40 mm, respectively. No light condition is conducive to produce spores for NH512, and the condition of light and dark 12 hours alternatives can promote spores for Y512, and the accumulation of spores are 9.184 × 107/cm2 and 9.038 × 107/cm2 for both strains on the 15 day. UV mutagenesis can induce mycelium growth of NH512, but inhibite Y512. UV mutagenesis can inhibite NH512 to produce spores, but induce to produce spores for Y512 in which the average accumulation of spores are 15.69 × 107/cm2.
