樟叶越桔不定根培养体系建立

Establishment of Adventitious Root Culture System of Vaccinium dunalianum

  • 摘要: 以樟叶越桔组培苗叶片为试验材料,分析不同暗培养时间、激素、碳源以及培养基等因素对不定根诱导的影响,建立了樟叶越桔叶片诱导不定根的培养体系,并进一步采用正交试验设计筛选出樟叶越桔叶片直接诱导不定根的最优诱导方案。结果表明:WPM + IBA 1.0 mg/L + 50.0 g/L白砂糖+暗培养15 d为不定根的最适诱导条件, 其生根率达96.7%。

     

    Abstract: Using the healthy leaves of tissue culture seedling of Vaccinium dunalianum as the material to directly establish the adventitious root culture system. The effects of different dark culture time, plant growth regulator concentration, carbon source and medium on the induction of adventitious root were investigated, and its induction condition was optimized by orthogonal design. The results showed that the optimal culture condition for leaf adventitious root induction was WPM + 1.0 mg/L IBA + 50.0 g/L sucrose with dark culture for 15 d, with a rooting rate of 96.7%.
