泡桐丛枝病发生过程中的蛋白质组学研究

Proteomic Study on the Occurrence of Paulownia Witches′ Broom Disease

  • 摘要: 为鉴定与泡桐丛枝病发生密切相关的蛋白质, 以毛泡桐健康苗、患丛枝病苗、20 mg/L或60 mg/L甲基磺酸甲酯(MMS)处理的丛枝病苗为实验材料, 用同位素标记相对与绝对定量(iTRAQ)技术检测毛泡桐幼苗4个样品的蛋白质表达变化, 并进行比对分析。结果表明:在鉴定到的1 622个蛋白质中, 共得到35个与泡桐丛枝病发生相关的差异表达蛋白质。在这35个差异表达蛋白中景天庚酮糖-1, 7-二磷酸酶(SBPase), 质膜ATP酶和光合系统Ⅱ 10 kDa多肽等蛋白质表达差异显著, 且主要参与了光合生物碳固定、光合作用和氧化磷酸化等生物学过程。

     

    Abstract: To identify the changes of proteins related to Paulownia witches′ broom (PaWB) disease occurring, isobaric tags for relative and absolute quantitation (iTRAQ) technique was employed to analyze the changes of proteins abundance among healthy Paulownia tomentosa seedlings, phytoplasma-infected seedlings, and the ones treated with 20 or 60 mg/L methyl methane sulfonate (MMS).The results indicates that a total of 1 622 proteins were identified, of which, 35 differentially abundant proteins were considered to be closely associated with PaWB disease.Among them, the expression of chloroplast sedoheptulose-1, 7-bisphosphatase (SBPase), plasma membrane ATPase, and photosystemⅡ10 kDa polypeptide were significantly differentially abundant.These proteins are mainly involved in carbon fixation in photosynthetic organisms, photosynthesis and oxidative phosphorylation.
