樟叶越桔VdARP7基因片段克隆与表达分析

Gene Cloning and Expression Analysis of VdARP7 in Vaccinium dunalianum

  • 摘要: 基于樟叶越桔叶芽转录组测序数据,筛选获得ARP7基因cDNA片段,采用RACE-PCR技术克隆了樟叶越桔VdARP7基因cDNA序列,并应用半定量PCR技术分析了VdARP7基因在樟叶越桔不同组织中的表达特征。结果表明:克隆了VdARP7基因964 bp cDNA片段,包括3′ UTR完整区域和PolyA尾巴特征,VdARP7属于Actin超级家族ARPs亚家族基因新成员,与多种高等植物已知ARP7基因具有较高的同源性,推导编码VdARP7蛋白的C-末端225个氨基酸残基序列,含1个NBD_sugar-kinase_HSP70_actin保守结构域。VdARP7基因能在樟叶越桔叶芽、嫩叶、老叶、花芽和花等组织中稳定表达,推测ARP7作为组成型表达的蛋白质在植物生长和发育等过程中发挥重要生理功能。

     

    Abstract: The cDNA fragment of ARP7 gene was isolated by transcriptome sequencing method from leaf bud of Vaccinium dunalianum. The cDNA sequence of VdARP7 gene was cloned with RACE-PCR. Semi-quantitative PCR was used to analyze the expression of VdARP7 gene in different tissues of V. dunalianum. The findings indicate that gene VdARP7 belong to the Actin superfamily ARPs subfamily and had high homology with ARP7 genes from many other higher plants. The open reading frame encoded a putative C-terminal sequence with 225 amino acid residues of VdARP7 protein. The VdARP7 also had the NBD_sugar-kinase_HSP70_actin conserved domain. The VdARP7 mRNA was stably expressed in leaf bud, young leaf, old leaf, flower bud and flower of V. dunalianum.It is speculated that ARP7 as a constitutively expressed protein plays an important physiological functions in plant growth and development.
