5个欧美杨无性系抗寒性测定与评价

Determination and Evaluation of Cold Resistance of 5 Populus×euramericana Clones

  • 摘要: 为比较引进欧美杨无性系抗寒性,以其1年生休眠苗为试验材料,采用人工梯度降温法在−15、−20、−25、−30、−35 ℃下处理24 h,测定其相对电导率、超氧化物歧化酶活性、过氧化物酶活性、过氧化氢酶活性及丙二醛含量、可溶性蛋白含量的变化,利用拟合的Logistic方程求出半致死温度,依据隶属函数法及相关性分析对测定指标进行评价和抗寒性排序。结果表明:低温胁迫下,酶活性、丙二醛含量及可溶性蛋白含量呈先升高后降低的趋势,无性系各指标间差异明显;无性系Qg的半致死温度最低且隶属函数值最大,各抗寒指标综合表现良好,其抗寒性最强;保护酶活性间呈极显著相关,电导率、丙二醛含量、可溶性蛋白含量间存在显著相关关系,6项指标可为杨树抗寒性研究测定提供参考依据。根据半致死温度及隶属函数值对抗寒性的排序为Qg>Ta>Pa>Ti>La>中林46>I-107。

     

    Abstract: In order to compare the cold resistance of introduced Populus×euramericana clones, the annual dormant seedlings were used as experimental materials, and the relative conductivity was determined by artificial gradient cooling method at −15, −20, −25, −30, −35 ℃ for 24 hours. The activities of SOD, POD, CAT and the changes of malondialdehyde and soluble protein content were measured under the low temperatures, and use the fitted logistic equation to find the semi-lethal temperature. The evaluation indexes and cold resistance were ranked according to the membership function method and correlation analysis. The results showed that under low temperature stress, enzyme activity, malondialdehyde content and soluble protein content increased first and then decreased, and the differences among different clones were obvious. The semi-lethal temperature of clone Qg was the lowest and the value of membership function was the largest. The cold resistance index has a good comprehensive performance, and its cold resistance is the strongest; the protective enzyme activity is extremely significantly correlated, and there is a significant correlation between conductivity, malondialdehyde content and soluble protein content. Six indicators can be used to determine the cold resistance of poplar and provide a reference basis. According to the semi-lethal temperature and membership function values, the order of cold resistance is: Qg>Ta>Pa>Ti>La>Zhonglin 46>I-107.
