Abstract:
On the basis of single-factor test, the effects of liquid to material ratio, compound enzyme addition and water bath temperature on the extraction of polyphenols from Yunnan distracted wood in
Juglans regia were analyzed. The optimal combination of polyphenol extraction from Yunnan distracted wood in
J. regia was determined by response surface design experiments with 3 factors and 3 levels. The Yunnan distracted wood in
J. regia polyphenol was purified by AB-8 macroporous resin with different concentration gradient of ethanol, and the suitable eluent concentration and the purity of polyphenols were analyzed. The findings indicated that under the conditions of liquid to material ratio of 102.00 mL/g, complex enzyme addition of 0.90% and temperature of 45.33 ℃, the extraction amount of Yunnan distracted wood in
J. regia polyphenols reached 22.35 mg GAE/g, and the actual value is 99.73% of the theoretical value; the value of Yunnan distracted wood in
J. regia polyphenols before purification was only 19.74%. After purification, the purity of Yunnan distracted wood in
J. regia polyphenols reached 64.42%, and the optimum ethanol concentration at the time of elution was 90%. Accordingly, by heating in water bath, the liquid to material ratio, the amount of complex enzyme added and the temperature of the water bath all have an effect on the extraction amount of polyphenols from the Yunnan distracted wood in
J. regia, and the elution degree of polyphenols in different concentrations of ethanol is different.
