西南远志中口山酮类化合物的生物活性研究

Study on Biological Activities of Xanthone Compounds from Polygala Crotalarioides

  • 摘要: 采用乙醇回流提取西南远志中的口山酮类化合物,利用AB−8大孔吸附树脂对其纯化,测定乙醇粗提物和纯化后总口山酮对黄嘌呤氧化酶的抑制活性;以DPPH自由基、ABTS自由基、O2•−、•OH清除率4种常用的体外抗氧化模型,分析其抗氧化活性;运用96孔板法对纯化后的口山酮5种细菌的抗菌活性进行实验,采用生长速率法测定分析纯化后西南远志中口山酮类化合物对5种真菌的抑制活性。结果表明:乙醇粗提物和纯化后总口山酮抑制能力的IC50分别为966、348.7 μg/mL,纯化后对黄嘌呤氧化酶抑制活性提高了2.77倍。对4种自由基都有一定的抗氧化能力,活性大小与浓度呈明显的线性关系,乙醇粗提物对DPPH自由基、ABTS自由基、O2•−、•OH自由基清除能力的IC50分别为283.85、226.37、304.36、236.82 μg/mL;纯化后总口山酮对DPPH自由基、ABTS自由基、O2• −、•OH清除能力的IC50分别为147.81、104.34、166.92、119 μg/mL。在对细菌的抑制活性方面,对变形杆菌表现出了最强的抑制作用,对金黄色葡萄球菌、铜绿假单孢菌、大肠沙门氏菌肠亚种的抑制能力次之,对大肠埃希氏菌的抑制能力最弱。在对真菌的抑制活性方面,对采绒革盖菌、腐皮镰孢、禾谷镰孢、尖孢镰孢、芸苔链格孢等5种真菌的EC50分别为0.237 9、0.507 4、1.184 4、0.899 1、0.368 7 mg/ml。

     

    Abstract: Ethanol reflux was used to extract xanthone compounds from Polygala crotalarioides, and AB−8 macroporous adsorption resin was used to purify xanthones. The inhibitory activity of xanthine oxidase by the crude ethanol extract and the xanthone compounds after purification was determined, DPPH, ABTS+•, O2•− and •OH clearance were used to study their antioxidant activities, the antibacterial activities of 5 kinds of bacteria were studied by 96-well plate method, the growth rate method was used to determine the inhibitory activities of 5 species of fungi. The results showed that the IC50 of the crude ethanol extract and the xanthone compounds after purification were 966 and 348.7 μg/mL, respectively. After purification, the inhibitory activity of xanthine oxidase was increased by 2.77 times. The IC50 of DPPH, ABTS+•, O2•− and •OH radical scavenging activities in ethanol extract were 283.85, 226.37, 304.36 and 236.82 μg/mL, respectively. After purification, the IC50 of DPPH, ABTS+•, O2•− and •OH radical scavenging capacity were 147.81, 104.34, 166.92 and 119 μg/mL, respectively. In terms of inhibitory activity against bacteria, it showed the strongest inhibitory effect on Proteus vulgaris. The second was the inhibition of Staphylococcus aureus, Pseudomanas aeruginosa and Salmonella enterica subsp. enteric. And the inhibition of Escherichia coli was the weakest. In terms of inhibitory activity against fungi, the EC50 of Coriolus versicolor, Fusarium solani, Fusarium graminearum, Fusarium oxysporum and Alternaria brassicicola were 0.237 9, 0.507 4, 1.184 4, 0.899 1 and 0.368 7 mg/ml, respectively.
