不同埋土深度杉木无性系萌蘖及内源激素含量的差异

Differences of Tillering and Endogenous Hormone Content of Cunninghamia lanceolata Clones in Different Soil Depth

  • 摘要: 以杉木‘洋020’扦插苗和‘洋020’组培苗为实验材料,通过盆栽实验,在调查不同埋土深度处理无性系萌蘖情况的基础上,应用酶联免疫吸附法测定了杉木无性系萌蘖高峰期枝叶、基部韧皮部、根尖等不同器官吲哚乙酸、赤霉素、细胞分裂素及脱落酸含量差异。结果表明:组培苗萌蘖能力强于扦插苗;随着埋土深度增加,2个无性系苗萌蘖能力均呈降低的趋势。不同埋土深度处理根尖和枝叶内源激素含量较高;随着培养时间延长,无性系枝叶吲哚乙酸含量呈上升趋势,赤霉素含量呈下降趋势;埋土深度6 cm处理有利于枝叶及根尖脱落酸的积累,埋土深度9 cm处理有利于根尖细胞分裂素的积累;无性系萌蘖与埋土深度呈极显著负相关,与赤霉素含量呈极显著正相关。不同埋土深度对杉木无性萌蘖有一定的影响,杉木无性系萌蘖高峰期均在8月份;不同器官植物内源激素含量是影响埋土深度处理杉木无性系萌蘖的主要影响因子之一。

     

    Abstract: Taking cuttings and plantlets of Cunninghamia lanceolata'Yang 020' as experimental materials, a pot experiment was conducted to investigate the effects of different soil depths on the tillering of C. lanceolata clones, the differences of indoleacetic acid, gibberellin, cytokinin and abscisic acid in different organs such as branches, leaves, basal phloem and root tip were determined by enzyme-linked immunosorbent assay. The results showed that the tillering ability of C. lanceolata'Yang 020' plantlets was stronger than that of tillering 'Yang 020' cuttage, and the tillering ability of 2 clones decreased with the increase of soil depth. The content of endogenous hormones in the root tip and the branches and leaves was higher under different soil depth treatment; the content of indoleacetic acid and gibberellin in the branches and leaves increased and decreased with the extension of culture time; the content of abscisic acid in the branches and leaves and the root tip was increased under 6 cm soil depth treatment, and the content of cytokinin in the root tip was increased under 9 cm soil depth treatment; there was a very significant negative correlation between clonal tillering and soil depth, and a very significant positive correlation between clonal tillering and gibberellin content. Different depth of buried soil had certain effect on the clonal tillering of C. lanceolata, and the peak of which was in August; the content of endogenous hormones in different organs was one of the main factors affecting the clonal tillering of C. lanceolata.
