芳樟叶片转录组测序及萜类合成调控相关基因表达分析

Transcriptome Sequencing of Linalool Leaf and Expression Analysis of Genes Involved in Terpene Synthesis

  • 摘要: 通过Illumina HiSeqTM 2000高通量测序技术对芳樟和杂樟(对照)叶片进行转录组学测序分析,并对所涉及的Unigene在GO、Nr、Nt、Pfam、KOG、Swiss−prot和KEGG中进行分类、功能注释和Pathway注释。结果表明:2类樟树叶片转录组检测共产生34.45 GB clean数据,组装了312 457个Unigene。其中,11 685个Unigenes鉴定为芳樟叶和杂樟叶的差异表达基因,在芳樟中6 481个被鉴定为上调的Unigenes和5 204个为下调Unigenes;对其进行GO功能富集发现并鉴定了编码萜类合成酶Unigenes 39个,包括单萜生物合成6个,二萜生物合成7个,倍半萜和三萜生物合成12个,萜类骨架合成14个。对萜类合成酶TPS14基因进行转录组FPKM和q−PCR验证发现,其表达量在芳樟中较杂樟更低,可能与其转录过程的选择性剪切有关。通过SSR位点分析,从88 016个Unigene中共检测到134 851个SSR位点,其中单个SSR位点有103 478个Unigene,含2个及2个以上SSR位点有31 373个Unigene。

     

    Abstract: In this study, Illumina HiSeqTM2000 high-throughput sequencing technology was employed to perform transcriptomes of linalool- and hetero-type camphor leaves. Classification, functional annotations and Pathway annotations were performed using GO, Nr, Nt, Pfam, KOG, Swiss-prot and KEGG databases. The results indicate that a total of 34.45 GB clean data was generated from 2 chemotypes of camphor leaves, and assembled into 312 457 Unigenes. Among them, 11 685 Unigenes were identified to be differentially expression in linalool- and hetero-type camphor leaves, including 6 481 up-regulated and 5 204 down-regulated Unigenes; 39 genes encoding proteins involved in terpene synthases, including 6 monoterpene biosynthesis, 7 diterpene biosynthesis, 12 sesquiterpene and triterpene biosynthesis, and 14 terpene skeleton synthesis. Three monoterpene synthase genes TPS14 were down-regulated in linalool-type compared to the hetero-type, and their expression were verified using transcriptome FPKM and q−PCR. A total of 134 851 SSR sites were detected from 88 016 Unigenes, including 103 478 Unigenes at single SSR sites and 31 373 Unigenes containing 2 and more SSR sites.
