南方红豆杉根际促生细菌蜡状芽孢杆菌CLY07的接种效应

Inoculation Effect of Growth-promoting Rhizobacteria Bacillus cereus CLY07 of Taxus chinensis var. mairei

  • 摘要: 以南方红豆杉根际土壤中分离筛选出的一株产ACC(1−氨基环丙烷−1−羧酸)脱氨酶的促生细菌CLY07菌株为研究对象,通过形态学、生理生化特征及16S rDNA系统发育分析对其进行鉴定,测定其产ACC脱氨酶、解磷、产IAA以及产嗜铁素等促生能力,并克隆其ACC脱氨酶相关基因acds。此外,将带有GFP标记的菌株接种于南方红豆杉幼苗根际,确定其在根际的定殖及动态;采用盆栽试验研究CLY07菌株对南方红豆杉实生苗的促生长作用。结果表明:该菌株被鉴定为Bacillus cereus,具有产IAA、产嗜铁素、溶磷等促生长特性,有促进植物耐盐碱的潜力,其ACC脱氨酶相关基因acds大小为996 bp;该菌株在植物根际及根表面能长期稳定定殖,在根际呈现先升后降,然后趋于平稳的规律,对南方红豆杉有明显促生长作用,能显著改善幼苗的生长状况,提高红豆杉的养分代谢能力。

     

    Abstract: Plant growth-promoting rhizobacteria strain CLY07 was studied, which was isolated from the rhizosphere soil of Taxus chinensis var. mairei and could produce ACC(1−aminocyclopropane−1−carboxylic acid) deaminase. This strain was identified by morphology, physiology-biochemistry, and 16S rDNA sequence analysis. Some of plant growth promoting characteristics such as 1−Amino Cyclopropan−1−Carboxylate(ACC) deaminase production, phosphate solubilizing capacity, production of auxin(IAA) and siderophore were evaluated. The ACC Deaminase Gene(acds) of strain CLY07 was cloned. A derivative of this strain, labeled with GFP, was also used to study the colonization of its hosts by pouring to the root surface of T. chinensis var. mairei. The effect of inoculation with this strain on the seedlings in pots was carried out. The results showed strain CLY07 was identified as Bacillus cereus, which could produce IAA and siderophore, showed phosphorus-solubilizing ability and salt tolerance. The full-length acds gene of strain CLY07 is 996 bp. This strain can colonize stably in the rhizosphere and root surface for long periods of time. At first, the number of the labelled cells rised, then decreased, and finally tended to be stable; Moreover, strain CLY07 can promote remarkably the seedling growth and nutrient metabolism of T. chinensis var. mairei.
