紫溪山华山松种子园无性系SCoT遗传多样性分析

SCoT Marker Analysis of Genetic Diversity in Pinus armandii Clones from Seed Orchard in Zixi Mountain

  • 摘要: 为明确云南省楚雄市紫溪山林场华山松种子园内不同种源无性系间的遗传背景,收集了园内6个种源的97份华山松无性系单株针叶,采用改良CTAB法提取其总DNA,并利用SCoT分子标记,对其进行遗传多样性分析,从而对园内华山松种质资源进行评价。结果表明:筛选出条带清晰、反应稳定且具有较高多态性的34条引物,经SCoT−PCR扩增后,华山松6个种源的等位基因数为5.2647,Shannon's多样性指数(I)为1.3786,Nei's基因多样性指数(H)为0.6904,说明华山松6个种源遗传多样性较高。地理位置较近的种源间其遗传距离并不一定较近,没有被归为一类,地理距离并未对其遗传距离造成明显影响。种源间的遗传分化系数(Gst)为0.0541,遗传变异主要存在于各种源内,占总变异的94.59%。华山松群体内基因流较大(Nm均值为4.3683>1),高水平的基因流可能削弱了各种源间的遗传差异。

     

    Abstract: In order to clarify the genetic background among the different provenances clones of Pinus armandii seed orchard in the Zixi mountain tree farm in Chuxiong City, Yunnan Province. The needles of 97 clones were taken from 6 provenances which was used to extract total DNA by modified CTAB method, and the genetic diversity was analyzed by SCoT molecular markers, to evaluate the germplasm resource. The main findings are as follows: 34 SCoT primers with clear bands and stable reaction were screened from the published SCoT primers in the existing literature. After SCoT-PCR were amplified, the allele number of 6 provenances of P. armandii was 5.2647, Shannon's information index(I) was 1.3786, Nei's gene diversity index(H) was 0.6904. It indicated that the genetic diversity of 6 provenances of P. armandii was high. The genetic distance between provenances in close geographical locations was not always close and were not in one category, and the geographical distance did not have significant influence on its genetic distance. The genetic differentiation coefficient among provenance(Gst) was 0.0541, and the genetic variation was mainly existed within each provenance, it accounted for 94.59% of the total genetic variation. The gene flow in P. armandii population was large(average Nm was 4.3683 > 1). So, high levels of gene flow may weaken genetic differences among various provenance.
