滇黄精多糖提取工艺及其抗氧化活性研究

Extraction Process and Antioxidant Activity of Polysaccharides from Polygonatum kingianum

  • 摘要: 以多糖提取率为指标,在单因素实验基础上,利用Box−Behnken 响应面法对滇黄精多糖提取条件进行优化;采用DEAE纤维素离子交换树脂纯化多糖后,以ABTS+·自由基的清除率体外抗氧化模型,测定纯化前后抗氧化活性。结果表明:滇黄精多糖的最优提取工艺参数为料液比1∶30(g/mL),提取时间1.5 h,提取温度80 ℃,滇黄精多糖提取率为20.70%。滇黄精粗多糖、纯化后中性多糖和酸性多糖清除 ABTS+·自由基的半清除浓度分别为 2.442、0.825、0.444 mg/mL,与样品浓度呈现一定的量效关系,且纯化后ABTS+·自由基清除率提高了5.55倍。

     

    Abstract: With the extraction rate of polysaccharide as the index, on the basis of single factor experiment, Box−Behnken response surface methodology was used to optimize the extraction conditions of polysaccharides from Polygonatum kingianum. The polysaccharides were purified by DEAE cellulose ion exchange resin. The antioxidant activity of polysaccharides before and after purification was determined by ABTS+· clearance in vitro model. The results showed that the optimum technological conditions were as follows: solid-liquid ratio: 1∶30(g/mL), extraction time: 1.5 h, extraction temperature: 80 ℃. Under these conditions, the extraction rate of PKP was 20.70%. The SC50 of crude polysaccharides, purified neutral polysaccharides and acid polysaccharides were 2.442, 0.825 and 0.444 mg/mL, respectively, which showed a dose-effect relationship with sample concentration, and the ABTS+· clearance was increased by 5.55 times after purification.
