寄主对桉树枝瘿姬小蜂体内可培养细菌多样性的影响

Effects of Different Hosts on the Diversity of Culturable Bacteria in Leptocybe invasa

  • 摘要: 为阐明桉树枝瘿姬小蜂对不同桉树的适应机制,采用传统的细菌分离与16S rDNA相结合的鉴定方法研究巨园桉(DH 201−2)和窿缘桉2种不同寄主对桉树枝瘿姬小蜂体内可培养细菌的影响。结果表明:以DH 201−2为寄主植物的桉树枝瘿姬小蜂体内可培养细菌的种类和多样性均高于以窿缘桉为寄主植物,其中以DH 201−2为寄主的桉树枝瘿姬小蜂体内可培养的细菌有4门6纲15属,菌群的丰富度指数、优势度指数、多样性指数和均匀度指数分别为0.90、0.84、2.03、0.72;以窿缘桉为寄主的桉树枝瘿姬小蜂体内可培养细菌仅有4门5纲9属,菌群的丰富度指数、优势度指数、多样性指数和均匀度指数也较低,分别为0.59、0.73、1.60、0.67。寄主为DH 201−2和窿缘桉的桉树枝瘿姬小蜂体内可培养优势菌属除了共有的泛菌属外分别为葡萄球菌属和短小杆菌属,说明寄主植物对桉树枝瘿姬小蜂体内细菌的种类、群落结构及多样性有显著影响。

     

    Abstract: In order to clarify the adaptation mechanism of Leptocybe invasa to different Eucalyptus, traditional isolation and which combined with 16S rDNA identification were carried out to isolate and identify the bacteria in the L. invasa adults parasitized DH 201−2(E. grandis × E. tereticornis) and Eucalyptus exserta, then the influence of different hosts on culturable bacteria in L. invasa could be find out. The findings indicated that L. invasa parasitize DH 201−2 hold a higher index in the variety and diversity of culturable bacteria, compairing with which parasitized Eucalyptus exserta. The culturable bacteria isolated from L. invasa parasitized DH 201−2 could be divided into 4 phyla, 6 classes and 15 genera. The index of richness, dominance, diversity and evenness of the bacteria from 4 phyla were 0.90, 0.84, 2.03 and 0.72, respectively. Bacteria isolated from L. invasa parasitized Eucalyptus exserta could be divided into 4 phyla, 5 classes and 9 genera. The index of richness, dominance, diversity and evenness of the bacteria from 4 phyla were 0.59, 0.73, 1.60 and 0.67, respectively, which were lower than the former. Apart from the common Pantoea, L. invasa both parasitized DH 201−2 and Eucalyptus exserta also had another 2 genera of vivo dominant bacteria, that is, Staphylococcus and Curtobacterium. These results indicated that host plants could significantly affect the species, community structure and diversity of bacteria in L. invasa.
