贵州省野生皂荚资源遗传多样性研究

Study on Genetic Diversity of Wild Gleditsia sinensis Resources in Guizhou Province

  • 摘要: 为探究皂荚资源的遗传多样性,采用相关系列扩增多态性(SRAP)分子标记技术对贵州省14个皂荚天然群体的203份样品进行了遗传多样性研究;通过聚类分析,探讨皂荚群体间Nei's遗传一致度、遗传距离分别与地理距离的相关性。结果表明:10对SRAP引物共扩增出77条清晰条带,多态性比例为98.70%;筛选出的SRAP 引物组合具有较好多态性,可用于皂荚的遗传多样性分析;观测等位基因数(Na)、有效等位基因数(Ne)、Shannon信息指数(I)和Nei's遗传多样性指数分别为1.7922、1.5613、0.4633和0.3174,皂荚居群间遗传分化系数(Gst)为0.2530,基因交流值(Nm)为1.4764,居群间存在较强的基因流,分化较小;群体遗传距离在0.0504~0.3596之间,平均值为0.162,地理距离与遗传距离呈显著正相关。14个群体可分为4个类群;其中黔南福泉单独作为一个类群。

     

    Abstract: To explore the genetic diversity of Gleditsia sinensis. The genetic diversity of 203 samples from 14 natural populations of G. sinensis in Guizhou Province was studied by using related series amplified polymorphism(SRAP) molecular marker technology. Cluster analysis was used to explore the correlation of Nei's genetic identity or genetic distance and geographical distance among G. sinensis populations. The results showed that 77 clear bands were amplified by 10 pairs of SRAP primers, and the polymorphism ratio was 98.70%. The selected SRAP primer combination had good polymorphism and could be used for the genetic diversity analysis of G. sinensis. The number of observed alleles(Na), effective number of alleles(Ne), Shannon information index(I) and Nei's genetic diversity index were 1.7922, 1.5613, 0.4633 and 0.3174. The genetic differentiation coefficient(Gst) among the G. sinensis populations was 0.2530, and the gene exchange value(Nm) was 1.4764. There was strong gene flow among the populations, and the differentiation was small; the population genetic distances ranged from 0.0504 to 0.3596, with an average of 0.162. Geographical distances were positively correlated with genetic distances. Cluster analysis showed that the 14 groups could be divided into 4 groups; among them, Fuquan in Qiannan was regarded as a group alone.
