不同光质对樟叶越桔组培苗生长及其6'−O−咖啡酰熊果苷含量的影响

Effects of different light quality on the growth of tissue culture seedlings of Vaccinium dualianum and the production of 6'-O-caffeoylarbutinfeoylarbutin

  • 摘要: 通过对樟叶越桔组培苗的生长生理指标及其中6'−O−咖啡酰熊果苷(CA)含量的测定,分析蓝色、红色、不同红蓝色配比(1∶1、1∶2和2∶1)及白色(对照)LED光对其生长和主要次生代谢物的影响。结果表明:单色红光处理可促进组培苗苗高的增长,蓝光处理可提高组培苗丙二醛(MDA)含量及过氧化物酶(POD)活性;红蓝光复合配比有利于其叶绿素含量和对光胁迫抗逆性的增加,在红蓝光比为1∶1时组培苗具有较高的超氧化物歧化酶(SOD)活性。当红光比例增加时,组培苗的可溶性糖及可溶性蛋白含量呈现先增加后减少的趋势;当蓝光比例增加时,组培苗的MDA含量和POD活性均为上升趋势;当红蓝光比为1∶2时,组培苗中CA的含量最高(21.02 ± 0.56) mg/g,为对照组的1.5倍,而在其他光质处理下其含量均下降。研究结果可为进一步阐明樟叶越桔CA的生产调控机制提供参考。

     

    Abstract: Light is a basic environmental factor in plant growth and development, which plays an important role in the regulation of plant growth and metabolism. In this study, the effect of red, blue, different ratios of red and blue (1∶1, 1∶2, and 2∶1), and white (control group) LED light on the growth and main secondary metabolites of tissue culture seedlings in Vaccinium dualianum was investigated by measuring the physiological parameters and the content of 6'−O−caffeoylarbutin (CA). The results showed that the growth of seedling height was promoted under the treatment of monochromatic red light, and MDA content and POD activity of seedling were enhanced under the treatment of monochromatic blue light. The chlorophyll content and resistance to light stress of seedling were increased under the treatment of combination of red and blue light. Of these, the tissue culture seedlings had a higher SOD activity when the ratio of red and blue light was 1∶1. The soluble sugar and soluble protein contents of tissue culture seedlings increased first and then decreased with the increase of red light proportion, while the MDA content and POD activity enhanced with the proportion of blue light increased. The highest content of CA in tissue culture seedlings was (21.02 ± 0.56) mg/g when the ratio of red to blue was 1∶2 with 1.5 fold higher compared to the control group, while its content decreased under other light-quality treatments. The results could provide an experimental basis and reference for the regulation mechanism of CA production in V. dunalianum.
