不同地理种源滇黄精内生菌的分离与功能研究

Isolation and Functional Exploration of Endophytes from Different Geographical Origins of Polygonatum kingianum

  • 摘要: 为探究滇黄精内生菌多样性组成及相关特性,以7个地理种源滇黄精根茎为研究材料,采用研磨稀释涂布法分离内生菌,并以平板拮抗、固氮、溶磷、解钾及促生实验对内生菌功能进行探索。结果表明:7个地理种源滇黄精根茎共分离到104株细菌、1株放线菌和1株真菌,大理下关的栽培种分离数量最多;内生细菌对禾谷镰刀菌、尖孢镰刀菌、链格孢霉菌、拟盘多毛孢菌和镰形炭疽菌具拮抗作用的分别有41、49、35、34、54株,T114、T115、T116、T117对多种病原菌均有拮抗效果;内生细菌中固氮菌有8株,效果较好的为D2−10、D2−5、D2−15菌株,解钾菌2株;104 cfu/mL的D2−5菌液对小麦生长有促进作用,种子萌发率、地上部分鲜质量、地上部分干质量、根长、根鲜质量比对照分别提高95.84%、54.35%、48.60%、35.96%、30.00%;D2−5菌株鉴定为Kosakonia cowanii。通过功能鉴定,初步筛选出一些具应用潜力的有益菌株。

     

    Abstract: To investigate the diversity composition and related characteristics of endophytes in Polygonatum kingianum, the endophytes were isolated by grinding, dilution and coating method from 7 geographical provenances. And the function of endophytes were preliminarily explored based on antagonistic plate bioassay, nitrogen-fixation, phosphate-solubilization, potassium-solubilization tests as well as seed germination-promotting experiments. The results indicated that a total of 104 strains of bacteria, 1 strain of actinomycete and 1 strain of fungus were obtained, of which the number of endophytes isolated from cultivated species in Dali Xiaguan was the largest. There were 41, 49, 35, 34 and 54 strains of antagonism to Fusarium graminearum, Fusarium oxysporum, Alternaria alternata, Polytrichosis pseudodiscoides and Colletotrichum falcatum, respectively. It should be noted that T114, T115, T116 and T117 have antagonistic effects on a variety of pathogens. Meanwhile, 8 bacterial strains could fix nitrogen, with D2−10, D2−5, and D2−15 strains showing better effects, and 2 strains solubilize potassium. In the growth promotion experiment of D2−5 strain, 104 cfu/mL bacterial solution has a promoting effect on the growth of wheat. The seed germination rate, aboveground fresh weight, aboveground dry weight, root length and root fresh weight increased by 95.84%, 54.35%, 48.60%, 35.96% and 30.00% respectively compared to the control. Based on morphological characteristics D2−5 strain was identified as Kosakonia cowanii. Through preliminary functional identification, some potential probiotics have been screened.
