2种栽培模式下紫皮石斛黄酮类物质差异分析
Differential analysis of flavonoids in Dendrobium devonianum under two cultivation modes

摘要: 以麻栎树3年生紫皮石斛为研究对象,利用超高效液相色谱串联质谱检测技术,对比分析2种栽培模式下紫皮石斛的化学成分。结果表明:麻栎树附生 3 年生紫皮石斛的总花青素含量高于人工设施栽培种植 3 年生紫皮石斛,但总黄酮含量较低;UPLC−MS/MS分析发现,栽培模式的改变对类黄酮的生物合成以及黄酮和黄酮醇的生物合成途径影响显著,其中根皮素、芹菜素、香橙素 (二氢山柰酚)、牡荆素−2″−O−葡萄糖苷等11 种差异代谢物含量随着栽培模式的改变呈上升趋势,上调物质主要与紫皮石斛的抗逆性有关,可以增加紫皮石斛中具有保护和抗逆作用的物质的合成;橙皮素、柚皮素*、柚皮素查耳酮*、3,5,7−三羟基黄烷酮 (短叶松素)等 13 种差异代谢物含量随着栽培模式的改变呈下降趋势,下调物质主要与抗氧化、抗炎、抗菌、抗纤维化、调控细胞凋亡等作用相关,表明紫皮石斛的成分与栽培模式密切相关。Abstract: Dendrobium devonianum is an epiphytic herb of the genus Orchidaceae. In order to study the effect of two cultivation modes on the quality of D. devonianum the chemical composition of D. devonianum under two cultivation modes was analyzed. The results showed that the total anthocyanin content of the 3-year-old epiphytic D. devonianum grown in the epiphytic of Jatropha curcas was higher than that of the 3-year-old D. devonianum grown in the artificial facility cultivation, but the total flavonoid content was lower. This suggests that the cultivation mode may affect the synthesis and accumulation of flavonoids in D. devonianum. Combined with UPLC-MS/MS technique analysis, it was found that the change of cultivation pattern had the most significant effect on the biosynthesis of flavonoids as well as the biosynthetic pathways of flavonoids and flavonols. Among them, 11 differential metabolites showed an upward trend with the change of cultivation pattern, including rhizopodophyllin, apigenin, chanterellin(dihydrokaempferol), and poncirin-2″-O-glucoside, etc. The up-regulated substances were mainly related to the resistance of Dendrobium, and could increase the synthesis of the substances with protective and antistress effects in Dendrobium. Thirteen differential metabolites showed a decreasing trend with the change of cultivation pattern, nerolidol, naringenin*, naringenin chalcone*, 3,5,7-trihydroxyflavanone(phragmitolone), etc. The down-regulated substances were mainly related to the effects of antioxidant, anti-inflammatory, anti-bacterial, anti-fibrotic, and regulation of apoptosis, etc. The down-regulated substances were mainly related to the antioxidant, anti-inflammatory, anti-microbial, anti-fibrotic, and apoptosis-regulating effects. These results indicate that the composition of D. devonianum is closely related to the cultivation mode, which is of great significance for further research on D devonianum.
