秋茄内生细菌群落结构特征与多样性研究
Structural Characteristics and Diversity of Endophytic Bacterial Communities in Kandelia candel

摘要: 为探究内生细菌与秋茄的互作关系,通过采集秋茄根、枝、叶组织样本,提取样本基因组DNA,采用16S rDNA高通量测序技术分析秋茄内生细菌群落结构与多样性,分析秋茄不同部位内生细菌群落的结构特征。结果表明:检测到26门、45纲、107目、186科、391属的内生细菌,共1893个OTUs。变形菌门在秋茄内生细菌群落中占据主体地位,甲基杆菌属是秋茄根、枝中的优势菌属,在秋茄叶中的相对丰度低于蓝细菌属。内生细菌群落丰富度呈现根 > 枝 > 叶,均匀度呈现枝 > 根 > 叶。优势菌群落与秋茄的新陈代谢、环境适应与遗传信息等功能显著相关。因此,秋茄根、枝、叶组织内生细菌群落的结构特征、多样性、优势菌属存在显著差异,内生细菌可能是秋茄具有优异适应性与抗逆性的重要因素。Abstract: To investigate the interaction between endophytic bacteria and Kandelia candel, root, branch, and leaf samples of K. candel were collected, followed by the extraction of genomic DNA from these samples. The community structure and diversity of endophytic bacteria within K. candel were analyzed utilizing 16S rDNA high-throughput sequencing technology. Additionally, the structural characteristics of endophytic bacterial communities across the various parts of K. candel were analyzed. The results showed that total of 26 phyla, 45 classes, 107 orders, 186 families, and 391 endophytic bacteria were detected, and the total number of OTU was 1893. Proteobacteria occupy the dominant position in the endophytic bacterial community, and Methylbacterium is the most predominant genus in the roots and branches of K. candel, and its relative abundance in leaves is lower than that of Cyanobacteria. The richness of endophytic bacterial communities were root > branche > leaf, and evenness were branch > root > leaf. The dominant bacterial community was significantly correlated with the functions of metabolism, environmental adaptation and genetic information. In conclusion, there were significant differences in the structural characteristics, diversity, dominant bacteria genera of the endophytic bacterial community in roots, branches and leaves, and endophytic bacteria may be an important factors for the excellent adaptability and stress resistance of K. candel.
