圆叶肿柄菊素A调控CYP724B1抑制番茄斑萎病毒NSmNSs基因复制研究

Tag A regulation of CYP724B1 to inhibite tomato spotted wilt orthotospovirus NSs gene replication

  • 摘要: 圆叶肿柄菊素A(Tag A)能抑制番茄斑萎病毒(TSWV)侵染,并通过转录组数据分析发现Tag A能诱导Nitab4.5_0007569g0010CYP724B1)、Nitab4.5_0001595g0020Nitab4.5_0004234g0120等10个基因的复制量变化。利用实时荧光定量PCR检测发现经Tag A处理后,Nitab4.5_0007569g0010CYP724B1)基因的复制量显著上调;利用病毒诱导的基因沉默(VIGS)技术,将CYP724B1基因沉默后进行预防实验处理。结果表明:基因CYP724B1沉默后,接种叶和系统叶中NSm、NSs基因的表达量与对照相比显著升高。将CYP724B1基因过表达后,接种叶和系统叶中NSm、NSs基因的表达量与对照相比显著降低。Tag A 通过调控CYP724B1基因抑制TSWV NSm、NSs基因的复制,进而抑制番茄斑萎病毒的侵染。

     

    Abstract: Tomato spotted wilt orthotospovirus (TSWV), a representative member of the Tospoviridae family. It causes serious damage to vegetables, such as tomato, pepper, pumpkin and celery. In previous study, the transcriptomic data analysis showed that Tag A could induce the expression of 10 genes (eg., Nitab4.5_ 0007569g0010 (CYP724B1), Nitab4.5_0001595g0020, Nitab4.5_0004234g0120)., Here, RT-qPCR was used to further verify the datas and found that the expression of Nitab4.5_0007569g0010(CYP724B1) gene was differently significant increased after Tag A treatment. CYP724B1 gene was silenced by virus-induced gene silencing (VIGS), the expression of the NSm and NSs genes were significantly increased compared to the control in the inoculated and systemic leaves. After CYP724B1 gene was overexpressed, the expression of the NSm and NSs genes were significantly decreased compared to the control in the inoculated and systemic leaves. These results indicate that Tag A could regulate the CYP724B1 gene to inhibit the expression of NSm and NSs to defense against TSWV infection.
