玉蕊花粉离体萌发及贮藏条件的筛选
Screening of optimal in vitro germination culture media and storage conditions of Barringtonia racemose pollen

摘要: 为筛选出适宜玉蕊花粉离体萌发的培养基以及玉蕊花粉贮藏的最适条件,以玉蕊的花粉为研究对象,采用不同浓度的蔗糖、H3BO3、CaCl2与不同水平的培养温度进行单因素和正交离体萌发试验。比较了醋酸洋红法、I2-KI (碘-碘化钾)法、TTC(氯化三苯四氮唑)法、MTT(二苯基四氮唑溴盐)法与离体萌发法测定花粉活力的结果。结果表明:玉蕊花粉活力的最佳测定方法为MTT法与离体萌发法,玉蕊花粉离体萌发的最佳培养基为蔗糖50 g/L、H3BO3 100 mg/L、CaCl2 25 mg/L,花粉萌发率为18.53%,培养温度为37 ℃。不同贮藏温度下的干燥处理与未干燥处理花粉萌发率均随着贮藏时间而逐渐降低,在−20 ℃且花粉经干燥处理的条件下,玉蕊花粉贮藏12后仍有萌发率。Abstract: In order to screen out the culture medium suitable for the in vitro germination of Barringtonia racemosa pollen and the optimal conditions for the storage of B. racemosa pollen, this study took the pollen of B. racemosa as the research object. Single-factor and orthogonal in vitro germination experiments were carried out by using different concentrations of sucrose, H3BO3, CaCl2 and different levels of culture temperature. The results of determining pollen viability by the acetic carmine method, I2-KI method, TTC method, MTT method and in vitro germination method were compared, and it was determined that the best methods for determining the viability of B. racemosa pollen were the MTT method and the in vitro germination method. The results showed that the optimal culture medium for the in vitro germination of B.racemosa pollen was sucrose at 50 g/L, H3BO3 at 100 mg/L, CaCl2 at 25 mg/L, and the pollen germination rate was 18.53%, with the culture temperature being 37 ℃. The germination rates of both dried and undried pollen under different storage temperatures gradually decreased with the storage time. Under the condition of −20 ℃ and after the pollen was dried, the B. racemosa pollen still had a germination rate after being stored for 12 units of time. The results of this study are of great significance for the population conservation, cross-breeding, and long-term preservation of germplasm of B. racemosa.
