油茶花粉生活力检测方法比较

Comparative Study on Pollen Viability Detection Methods for Camellia oleifera

  • 摘要: 以7个油茶种的花粉为试材,采用TTC法、无机酸法、离体萌发法、活体萌发法4种方法对其花粉生活力进行测定。结果表明:TTC法适合山茶属植物花粉生活力的检测,而无机酸法不适合;活体萌发法检测油茶花粉生活力不准确,但能有效表述花粉萌发力;离体萌发法能有效准确地测定油茶花粉生活力,适合山茶属植物花粉生活力的检测,是最为理想的快速测定法。

     

    Abstract: The pollen vitality of seven species in genus Camellia were determined by four different methods i.e., triphenyl tetrazolium chloride (TTC), mineral acid, in vitro and in vivo germination. The results indicated that TTC method, but not the mineral acid method, was suitable to detect the pollen viability of Camellia genus, The detect result obtained from in vivo germination method was not so accurate, but it could be applied to describe the pollen vigor more effectively. It was showed that the in vitro germination method was the best and the most ideal way to determine the pollen viability for species in Camellia genus.
