厚荚相思光自养微繁殖技术研究

Study on Techniques of Photoautrophic Micropropagation of Acacia crassicapa

  • 摘要: 以厚荚相思组培苗为材料进行光自养微繁殖的生根培养研究。结果表明:不同培养基质对厚荚相思的生根有显著影响,以河沙为最优;不同营养液对厚荚相思的生根也有显著影响,以不含营养成分的水分为最优;不同光强模式在透气性好的河沙和透气性差的琼脂培养基上,对厚荚相思的生根、地上生物量和地下生物量有显著影响,光强模式2(0~7 d低光强,光强为2500~3000 lx,光照时间12h/d;8~10d光强达到4000~5000lx,光照时间14h/d;10d以后光强要达6000~8000lx,光照时间14h/d)为最优。

     

    Abstract: A study on the rooting culture of the Acacia crassicapa plantlets through photoautotrophic micropropagation was carried out.The results showed that the different culture media had significant effects on the rooting process of A.crassicapa, of which river sand was the optimal medium.The different nutrient solutions also had significant effects on the rooting of A.crassicapa in the photoautotrophic micropropagation experiment, of which the pure water without any nutritional components was the most effective.Different light intensity modes had significant effects on the rooting process on the above ground biomass and the underground biomass of A.crassicapa, of which Mode 2 was the optimal, i.e.the low light intensity within the range of 2500-3000 lx with the illumination duration of 12 h/d from the 0 to the 7th day; while the intermediate light intensity in the range of 4 000-5 000 lx with the illumination duration of 14 h/d between the 8th to the 10th day; and the moderate light intensity in the range of 6 000-8 000lx with the illumination duration of 14 h/d after the 10th day was applied separately in the different stages.
